Summary:
This master thesis focuses on monitoring plasma metabolome changes during time
after wine consumption, unravelling compelling evidence of wine health effects and
bioavailability of phenolics present in wine. We specifically examined the metabolome
of human plasma samples after four nutritional interventions: over four separate days,
10 healthy participants consumed a standardized meal with either white wine (Robola),
red wine (Cabernet Sauvignon), ethanol solution, or water (cross-over study). Plasma
samples were analysed using Liquid Chromatography coupled to High Resolution
Mass Spectrometry (LC-QTOF/MS). A suspect screening protocol was followed using
an in-house suspect list of phenolic wine metabolites.
Chemometrics were implemented to enlighten the holistic metabolic perturbation in
response to acute red wine consumption in humans compared to changes induced
only by ethanol or white wine. Elucidation of time-dependent metabolome alterations
after acute wine consumption is, to our knowledge, studied for the first time.
