Electrophoretic and antigenic properties of turtle myoglobins

1. 1. Turtle myoglobins (Mbs) were isolated from skeletal muscles. Non-myoglobin proteins were removed and Mb further purified by cation exchange chromatography. 2. 2. By disc electrophoresis multiple Mbs were revealed. One major and four minor components were detected. All five turtles tested were found to contain Mbs showing similar electrophoretic patterns. 3. 3. Quantitative immunoprecipitation analysis with specific antisera showed that equivalence zone is reached with a Mb concentration corresponding to O.D. of 0·24 at 424 nm in a 10-mm curvette. 4. 4. Using antisera against Mbs from Clemmys caspica, the percentages of immunological cross-reactions of heterologous relative to homologous antigenic species, are: Emys orbicularis = 36·2, Testudo hermanni = 30·0 and Testudo graeca = 24·8. © 1973.
